Knotting is required only for good quality pearls, to keep them from damaging each other while rubbing together.
The knotted look can also be a design choice, or functional, as when making malas, or prayer strands, with spaces between beads to facilitate counting
Generally, silk is not considered durable enough to use for un-knotted necklaces, use fine soft cables like Flex-Rite instead.
For knotting it's usually best to use the largest size silk cord that will fit through all of your beads. If you are knotting different kinds of beads together, they should have similar sized holes. Test whether the cord will go thru all the types of beads your are using in the necklace before getting started.
Often a card of silk will be enough to knot two necklaces, so be sure to unwind the card and start knotting at the end without the needle. This way the piece you have left over will still have a needle attached to use on the next project.
Most gemstones will take size 6 or 8 silk cord
Some pearls and small faceted gems require smaller cords, size 4 or 2.
